Western Blot primary and secondary antibody stripping buffer (also known as membrane regeneration solution or antibody removal solution) is used for reusing membranes bound with antibodies in Western blot. This reagent allows the removal of bound antibodies so that the same membrane can be reprobed. Typically, a membrane can be reused 2–3 times, although repeated use may lead to weakened protein signals, making further reuse unsuitable.

Alkaline stripping buffers use high pH and detergents for strong antibody removal, best suited for durable PVDF membranes, though they may damage proteins on NC. They are ideal for complete stripping in ECL detection.

Acidic stripping buffers use low pH for gentler antibody removal, preserving proteins on both PVDF and NC membranes, making them suitable for multiple reprobings, though stripping may be incomplete.
